In the early days of microscope photography, we usually simply connected an ordinary 135 camera to the upright barrel of a microscope, and at the same time, when the camera lens was not removed, the microscope was performed in the same way as a general camera. photograph. But we should also pay attention to the following points:
* Point, the camera should be placed in an appropriate position above the eyepiece, so that the exit pupil is just on the front surface of the camera lens, the position of the exit pupil can be checked simply with a piece of fluorescent glass and a piece of white paper, when the microscope is connected to the digital camera, it Should be at the smallest diameter of the outgoing beam. And in addition to the light from the microscope, the cover should also prevent other light from entering the camera.
The second point is to focus the microscope image and adjust the distance on the camera to infinity, and the image will naturally fall on the film plane. At the same time, it is more convenient to use a single-lens reflex camera, and the microscope image can also be directly focused by the observation window of the camera.
The third point is that the aperture of the camera lens should also be selected to a larger value, a small aperture may reduce the illuminated area on the film plane, but it does not affect the overall brightness of the image.
Of course, it may be inconvenient to use an ordinary camera for photomicrography, but it can still be said to be a more effective method when there is no special microscopic camera.
