Sample preparation issues
1. Sample thickness
Confocal microscopy has strict requirements for sample thickness. Thick samples can cause light scattering and signal attenuation, affecting imaging quality. Therefore, it is necessary to control the thickness during sample preparation to ensure that the laser can effectively penetrate and return to the detector.
Solution: Optimize slicing techniques by using ultra-thin slicing or cell culture techniques to reduce sample thickness.


2. Sample staining
Staining is a crucial step in confocal imaging, but improper staining may result in weak signal or high background.
Solution: Choose appropriate fluorescent dyes, optimize dyeing conditions, including dyeing time, temperature, and pH value.


Laser and detector issues
1. Unstable laser power
The fluctuation of laser power can affect the consistency and repeatability of imaging.
Solution: Regularly calibrate the laser to ensure stable power and use high-quality lasers.


2. Insufficient sensitivity of the detector
Insufficient sensitivity of the detector may result in weak signals, making it difficult to obtain clear images.
Solution: Use a high-sensitivity detector or increase the laser power, but be careful not to damage the sample.


Imaging parameter setting issue
1. Inappropriate pinhole size
The size of the pinhole directly affects the resolution and depth of imaging. Pinholes that are too large or too small can lead to a decrease in image quality.
Solution: Adjust the pinhole size according to the sample characteristics and imaging requirements.


2. Scanning speed too fast or too slow
Scanning speed affects imaging time and image quality. Too fast may cause blurry images, while too slow can waste time.
Solution: Set the scanning speed reasonably according to the characteristics of the sample and the experimental purpose.


Image processing issues
1. Image artifacts
During confocal imaging, artifacts such as halos and stripes may occur.
Solution: Optimize imaging parameters, use high-quality optical components, and perform post image processing.


2. Insufficient dynamic range
Insufficient dynamic range of the image may lead to signal saturation or loss.
Solution: Adjust the laser power and gain, and use a high dynamic range camera.


Environmental factor issues
1. Temperature and humidity fluctuations
The changes in environmental temperature and humidity may affect the sample state and instrument performance.
Solution: Perform imaging in a constant temperature and humidity environment, or use temperature and humidity control devices.


2. Vibration interference
Vibration in the laboratory may affect imaging stability.
Solution: Use an anti vibration table or perform imaging during periods of low vibration.
