Difference between normal light microscope and fluorescence microscope
Optical microscope: An optical instrument that uses optical principles to magnify and image tiny objects that cannot be distinguished by the human eye, so that people can extract information about the microstructure.
Fluorescence microscope: Fluorescence microscope uses ultraviolet light as a light source to irradiate the object to be examined and make it emit fluorescence, and then observe the shape and location of the object under the microscope.
Fluorescence microscope: Fluorescence microscope uses ultraviolet light as a light source to irradiate the object to be examined, causing it to emit fluorescence. Fluorescence microscopy is used to study the uptake and transport of substances, distribution and localisation of chemicals in cells. Some substances in the cell, such as chlorophyll, can fluoresce after irradiation by ultraviolet light; some other substances itself can not fluoresce, but if stained with fluorescent dyes or fluorescent antibodies, fluorescence can also fluoresce by ultraviolet light irradiation, fluorescence microscopy is one of the qualitative and quantitative study of such substances tools.
Optical microscopes are generally composed of a stage, a spotlight illumination system, an objective lens, an eyepiece and a focusing mechanism. By changing the illumination method, the spotlight illumination system can obtain different observation methods, such as dark object points on a bright background (known as bright field illumination) or bright object points on a dark background (known as dark field illumination), so as to better discover and observe microstructures under different circumstances.
Fluorescence microscope and ordinary microscope have the following differences.
1. The illumination method is usually drop-type, i.e. the light source is projected on the sample through the objective lens.
2. The light source is ultraviolet light, which has a shorter wavelength and a higher resolving power than ordinary microscopes.
3. There are two special filters, the one before the light source is used to filter out visible light, and the one between the eyepiece and the objective lens is used to filter out ultraviolet light to protect the human eye.
Fluorescence microscope is also a kind of optical microscope, the main difference is that the excitation wavelength of the two are different. The main difference is that the excitation wavelength of the two is different, which determines the difference in the structure and use of fluorescence microscope and ordinary optical microscope.
Fluorescence microscope is the basic tool for immunofluorescence cytochemistry. It is composed of light source, filter plate system and optical system and other major components. It uses a certain wavelength of light to stimulate the specimen to emit fluorescence, which is magnified through the objective lens and eyepiece system to observe the fluorescence image of the specimen.
