How to adjust the laser intensity of a confocal laser scanning microscope

 

Power control knob settingNormally set it to 30% of the maximum value. Higher output shortens laser service life and causes laser noise. Adjust AOTF and PMT first to obtain optimal intensity.

 

Acousto-optic tunable filter functionIt controls excitation beam and intensity. It switches laser within milliseconds and adjusts intensity from 0% to 100%. Different laser parameters can be applied to each image point.

 

Working principleIt adopts Bragg diffraction effect of sound waves in medium. Input radio frequency signal makes AOTF diffract polychromatic light and select specific monochromatic light. Wavelength corresponds to radio frequency. Electric tuning realizes fast wavelength switching. AOTF supports continuous laser adjustment, local stimulation scanning and microsecond-level switching.

 

In confocal laser microscope, PMT cannot increase laser intensity but adjusts imaging signal strength. Adjusting PMT voltage changes image brightness. Confocal pinhole size also influences brightness. PMT is sensitive to ultraviolet, visible and near-infrared light and amplifies weak light signals for measurement.

 

Maintenance of metallographic microscope
1. The laboratory environment should have three protective conditions: earthquake resistance (away from the earthquake source), moisture resistance (using air conditioning and dryers), and dust prevention (laying a floor on the ground); Power supply: 220V+-10%, 50HZ Temperature: 0 ° C-40 ° C.

 

2. Do not switch the objective lens when the center of the circular hole on the stage pad is far away from the center of the objective lens, in order to avoid scratching the objective lens.

 

3. When focusing, be careful not to let the objective lens touch the sample to avoid scratching the objective lens.

 

4. All function switches should be light and precise.

 

5. When adjusting brightness, it is important to avoid sudden changes in brightness or excessive brightness, as it can affect the lifespan of the light bulb and also damage vision.

 

6. When replacing halogen lamps, pay attention to high temperatures to avoid burns. Do not directly touch the glass body of the halogen lamp with your hands.

 

7. Place infrequently used optical components in a drying dish.

When turning off the metallographic microscope, adjust the brightness to * low before turning it off.

 

9. Non professionals should not adjust the lighting system (filament position lamp) to avoid affecting the imaging quality.

 

10. When shutting down, adjust the objective lens to the * low state through the focusing mechanism, and cover it with a dust cover after cooling. Pay attention to fire prevention.

 

11. Non professionals should not attempt to wipe the objective lens and other optical components. The eyepiece can be wiped with a degreased cotton swab dipped in a 1:1 mixture of liquid and shaken dry. Do not use other liquids to avoid damaging the eyepiece.
