How to operate an inverted microscope correctly?

 

1. The most commonly used observation method in an inverted microscope is phase contrast. Since this method does not require staining, it is ideal for observing living cells and microorganisms. A variety of concentrators are offered here to suit needs, this method provides high contrast, high definition images with natural background colours.


2. Power on. Connect to power. Turn on the electric control switch at the lower end of the mirror body.

(1) Preparation: Place the object to be observed on the stage. Rotate the three-hole converter to select the smaller objective. Observe and adjust the hinged binocular eyepieces for comfort.


(2) Adjust the light source: Push and pull to adjust the brightness adjuster at the lower end of the mirror body to the appropriate level. Adjust the size of the light source by adjusting the grating under the condenser.


(3) Adjust the image distance: turn the three-aperture converter to select an objective lens with a suitable multiple; replace and select a suitable eyepiece; at the same time adjust the lift to eliminate or reduce the halo around the image and improve the contrast of the image.


(4) Observation: Observe the results through the eyepiece; adjust the stage and select the observation field.

Remove the observation object, and push and pull the brightness adjuster of the light source to the darkest. Turn off the switch at the lower end of the mirror body and disconnect the power supply. Rotate the three-hole converter so that the objective lens is placed on the lower side of the stage to prevent dust from settling.


Daily maintenance and precautions


1. All lens surfaces must be kept clean. The dust that falls on the lens surface can be blown off with an ear suction ball or gently dusted off with a soft brush.


2. When the surface of the lens is stained with oil or fingerprints, use absorbent cotton dipped in a little mixture of absolute ethanol and ether (3:7) to wipe it gently.


3. Do not use organic solution to clean the surface of other parts, especially plastic parts, use a soft cloth dipped in a small amount of neutral detergent to clean.


4. Under no circumstances should the operator wipe the lens surface with cotton balls, dry cloth or dry lens paper, otherwise it will scratch the lens surface and seriously damage the lens, and do not wipe the lens with water, as this will leave some water stains, which can cause mold growth and serious damage to the microscope.


5. During the intermittent period of instrument operation, in order to prevent dust from entering the lens barrel or lens surface, the eyepiece can be left on the lens barrel, or covered with a dust-proof plug, or the instrument can be covered with a dust-proof cover.


6. The micromirror should not move as much as possible. If it needs to be moved, it should be handled with care to avoid collision.


7. It is not allowed to disassemble the instrument at will, especially the intermediate optical system or important mechanical parts, so as not to reduce the performance of the instrument.
