How to use phase contrast microscope
1. Swap and install the phase difference device. Remove the condenser used in ordinary microscopes, install the annular aperture on the condenser bracket, and place the green filter on top. It can absorb red and blue light, illuminating monochromatic light with a small wavelength range and having a heat absorption effect, which can achieve good results in phase difference observation. Then unscrew the regular objective lens from the converter and replace it with a phase difference objective lens.
2. Focus and turn on the light source, rotate the concentrator dial, align the "o" with the marked hole, and allow the ordinary concentrator part to enter the light path. First, use a low contrast objective lens and follow the normal microscope operation method to adjust the light and focus. Rotate the annular aperture so that the diameter and aperture width of the aperture are compatible with the phase contrast objective used. For example, when the phase contrast objective is 40x, use x40 to indicate the aperture of the aperture.
3. Adjust the uranium and remove the eyepiece, insert the uranium telescope, observe from inside the telescope, and fix the outer tube with the left hand; Rotate the inner cylinder of the telescope with your right hand to lower it, and when it aligns with the focus, you can see the bright ring of the circular aperture and the black ring of the phase plate. At this point, you can fix the telescope. Then raise and lower the light collector and adjust the spiral below it to make the size of the bright ring consistent with the black ring, and then adjust the adjustment knob on the annular diaphragm condenser from left to right, back and forth to make the two rings coincide completely. If the bright ring is smaller than the black ring and located inside, lower the light collector to enlarge the bright ring; On the contrary, the concentrator should be raised to reduce the brightness of the ring. If it cannot completely overlap even after reaching the maximum limit, it may be due to the glass slide being too thick and should be replaced. After the adjustment is completed, remove the telescope, switch back to the sight glass, and observe according to the usual procedures. When replacing phase difference objectives with different magnifications, it is necessary to use a matching annular aperture and readjust the aperture each time. When using an oil mirror, asphalt should be added between the surface of the lens on the collector and the glass slide at the same time.
