Instructions for using micrometers:

The micrometer is divided into an eyepiece micrometer and a mirror micrometer, which are used together. The eyepiece micrometer is a circular piece of glass with a central notch of one foot, measuring 5-10mm in length and divided into 50-100 grids. The actual length represented by each varies depending on the magnification of different objective lenses and the length of different lenses. A mirror micrometer is a circular side micrometer located in the center of a glass slide, sealed with gum, measuring 1-2 mm in length and divided into 100 or 200 grids. The actual length of each grid is 0.01mm (10 μ m) . When using an eyepiece micrometer to measure the size of cells, it is necessary to first verify the actual length represented by each grid of the eyepiece micrometer using a stage micrometer.


The method is as follows:

1. Pull one side of the eyepiece out of the lens barrel, unscrew the part below the eyepiece, place the micrometer scale on the focal plane of the eyepiece downwards, reinstall the unscrewed part back into the eyepiece, and then insert the eyepiece back into the lens barrel.


2. Place the micrometer scale on the mirror stage upwards and clamp it so that the micrometer scale is located in the center of the field of view. Focus until you can clearly see the indexing of the micro ruler on the side of the mirror.


3. Carefully move the micro ruler on the mirror table side and rotate the micro ruler on the eyepiece side to make a straight line to the left of the two rulers coincide, and then find the straight line that coincides again from left to right.


4. Record the number of grids between the eyepiece side micrometer and the mirror table side micrometer between two overlapping lines. Calculate the actual length represented by each grid of the eyepiece side micrometer. The actual length represented by each grid of the eyepiece micrometer=(the number of grids of the stage micrometer between the two merging lines/the number of grids of the eyepiece micrometer between the two merging lines) × 10um5. Remove the mirror side micrometer, replace it with the glass slide specimen that needs to be measured, and use the eyepiece side micrometer to measure the standard.
