Introduction to the use of microscope micrometers
The microscope is measured with a micrometer engraved with a certain scale. First, the objective lens micrometer with a length is used to correct the eyepiece micrometer that does not indicate the length, and the actual length represented by each grid of the latter is calculated, and then the specimen to be measured is placed. , use the eyepiece micrometer to measure the number of grids of the eyepiece micrometer occupied by the structure on the specimen, and then calculate the size of the cell.
Microscope eyepiece micrometer is a circular glass slide, usually 5mm is divided into 50 grids, each grid is actually equal to 100μm. It must be calibrated with an objective micrometer before use.
Olympus Microscope Objective Micrometer A special slide with a small circle in the center. There are graduations engraved in the circle, and the straight line with a length of 1 mm is divided into 100 small divisions, and each division is equal to 10 μm.
Calibration of the eyepiece micrometer
(1) Take off the eyepiece, unscrew the eyepiece on the eyepiece, put the eyepiece micrometer on the middle partition of the eyepiece, make the side with the scale face down, then screw on the eyepiece, and put it into the lens barrel Inside.
(2) Place the objective micrometer on the stage of the microscope, so that the scaled side faces up, and the small circle with the scale is located in the center of the field of view, just like observing the specimen.
(3) Observe with a low-power lens first, align the focal length, and after seeing the scale of the objective lens micrometer clearly, turn the eyepiece so that the scale of the eyepiece micrometer is parallel to the scale of the objective lens micrometer, and make the two-foot scale The first line on the left coincides, and then look for another coincident line two feet to the right.
