Operating Methods for MEIJI Microscopes
(1) Inspection: Grasp the microscope arm with your right hand, take the microscope out of its case, support the base with your left hand, and place it gently on the laboratory bench. First inspect all microscope components for damage. If any damage or malfunction is found, report it to the instructor immediately for handling.
(2) Preparation: Place the microscope slightly to the left of your front side. Tilt the arm if necessary (some microscopes are pre‑tilted) for easier observation. Turn the coarse adjustment knob to slightly raise the tube (or lower the stage) to increase the clearance between the objective lens and the stage. Then rotate the objective turret to align the low‑power objective with the aperture in the center of the stage (a click sound can be heard).
(3) Light Alignment: Open the diaphragm and raise the condenser. Keep both eyes open; look into the eyepiece with your left eye while adjusting the angle of the mirror until the field of view is brightly and evenly illuminated. The plane mirror tends to reflect surrounding objects into the view, so the concave mirror is generally used for light alignment.
(4) Mount the Specimen Slide: Place the coverslip side of the specimen slide upward. Set the slide at the front of the stage and push it under the objective lens, then secure it with the stage clips. If a mechanical stage is fitted, fasten the slide with its spring clip, and move the target area to the center of the aperture.
(5) Focus Adjustment: Watch the objective lens from the side of the microscope while turning the coarse adjustment knob to slowly lower the tube (or raise the stage). Stop when the gap between the low‑power lens and the slide is approximately 5 mm. Then look through the eyepiece with your left eye, and slowly turn the coarse adjustment knob with your left hand to raise the tube until the specimen image appears in the field of view. If the image is not sharp enough, turn the fine adjustment knob to achieve clearer focus.
