Precautions and maintenance of fluorescence microscope
(1)Operate in strict accordance with the requirements of the fluorescence microscope factory manual, do not change the procedure at will.
(2)It should be examined in the dark room, turn on the power supply of the high voltage regulator after entering the dark room, turn on the excitation high pressure mercury lamp after turning it on for 3~5rain, and then turn on the excitation high pressure mercury lamp, and stop the excitation when you see that the high pressure mercury lamp is completely bright, then start observing the specimen.
(3) Prevent UV damage to the eyes, put down the protective screen before doing the experiment.
(4) Check the time each time to 1h is appropriate, more than 90min, high-pressure mercury lamp luminous intensity gradually decreased, fluorescence weakened, the specimen was excited 15min, fluorescence is also significantly weakened.
(5) fluorescence microscope excitation device and high-pressure mercury lamp life is limited, the specimen should be centralised inspection, save time; mercury lamp should avoid multiple open, * good to avoid switching within half an hour.
(6)Observe the specimen immediately after staining, because the storage time is too long, the fluorescence will gradually burst. Can be stained specimens wrapped in black paper, stored in polyethylene plastic bags, 4 ~ C storage, can delay the fluorescence burst time.
(7)The judgement standard of fluorescence brightness is generally four levels, i.e. 'a' no or visible weak autofluorescence; '+' only visible fluorescence; '++' visible bright fluorescence. fluorescence; '+++' visible dazzling fluorescence.
Precautions and maintenance of fluorescence microscope:
1. Lens cleaning dust stuck on the lens should be soft brush gently brush off, in the place of fingerprints and oil stains should be soft in the net of degreasing cotton, gauze or mirror paper dipped in anhydrous ethanol (or methanol) gently wipe, on the surface of the objective lens can be used to wipe the oil stains with gasoline.
