Precautions for Microscopic Image Acquisition with a Microscope
To obtain high-quality microscopic images / capture micrographs with a microscope, you must follow standard operating procedures. The key points listed below are essential:
① When using the microscope, check the specimen to be observed carefully: confirm whether the section is too thick and whether the coverslip meets the specifications.
② Objective lenses of the microscope are generally available in 6–8 magnification steps. Plan achromatic objectives are commonly used. If the objective magnification is too low, the image quality will degrade. Therefore, it is recommended to select a plan objective of an appropriate magnification.
③ Adjust the condenser aperture diaphragm. Match it with the numerical aperture of the objective lens to achieve optimal image resolution.
④ The focusing system is a critical mechanical component, and proper focusing operation is very important. For beginners in biology or new microscope users, proficient operation of the focusing system helps quickly locate and focus on the specimen, improving work efficiency.
⑤ When observing specimens on the stage, avoid frequent X-Y movement. A double-layer mechanical stage offers good precision and stability, which can provide high-quality images during microscopic observation.
⑥ An aspheric condenser lens can provide highly uniform illumination, so that the observed microscopic images have good contrast.
⑦ The field diaphragm should be opened to an appropriate position according to the observed specimen and objective magnification. If opened excessively or insufficiently, the brightness of the microscopic field will be affected.
⑧ If the illumination system adopts a low-heat, high-color-temperature lighting device, microscopic images with high resolution and sharpness can be observed.
⑨ The Kohler illumination system can adjust the optical center. Research-grade microscopes are equipped with this system, which can provide high-quality micrographs for microscopes and professional digital microscopic imaging systems.
