Self-calibration method of biological microscope
1 Purpose
This method specifies the self calibration method for biological microscopes, ensuring that the self calibration of biological microscopes is carried out correctly according to the prescribed method.
2 Scope of application
This method is applicable for the self calibration of biological microscopes used and repaired in our center.
3 Responsibilities
3.1 The instrument responsible person shall strictly follow this method, calibrate on schedule, keep records, and issue calibration reports.
3.2 The department head reviews the calibration results.
3.3 The technical leader issues the calibration report.
3.4 The equipment department will archive a copy of the calibration report.

5 Overview
6 Technical Requirements
6.1 Instrument Appearance
The eyepiece, objective lens, condenser lens and other components are undamaged.
The moving and rotating parts of the microscope should have a comfortable feeling and should not be too tight or loose.
6.2 Optical System Imaging Quality
Turn on the light source, reduce the aperture of the diaphragm, and verify the degree of coincidence between its center and the center of the aperture of the condenser, objective lens, and eyepiece.
Observe the star spot plate from low magnification to high magnification, and verify the spherical aberration, chromatic aberration, coma, and astigmatism of the objective lens.
6.3 Changing the magnification of the mechanical mirror
Rotate the objective lens converter to change the magnification of the objective lens and observe the center defocus of the image plane.
6.4 Scale and lettering
All scales and inscriptions on the microscope must be clear and distinct.
6.5 Verification cycle
The biological microscope has a one-year calibration cycle.
7 Environmental conditions for calibration
Temperature: 25+2 ℃, humidity: 45%+2%
8. Main measuring equipment used for calibration
Star point board, measurement range: objective lens (0-1000) X, * maximum allowable error: 0.1%.
