Steps for observing bacteria with a microscope
Make tablets using bacteria from dirty hands:
1. Collect bacteria on hands, rinse dirty hands with a small amount of sterile water (cold boiled water can be used instead), and let the handwashing water flow into the petri dish. This dirty water is the bacterial culture fluid.
2. To prepare bacterial mounting slides, take two clean slides A and B, drop a small drop of bacterial culture solution in the center of each slide; then cover slide A with a cover glass and directly use 600 times microscope observation.
(Use a slightly darker field of view and adjust it properly to see a variety of bacteria and other microorganisms. It is easy to see active microorganisms, which is more attractive and educational for children.) If you want to further zoom in and know how to operate, select After determining the object to be observed, you can move the object to the center of the field of view, and then switch to a higher magnification lens for observation.
3. Put a small drop of gentian violet staining solution on the bacterial culture solution on slice B. Use a toothpick to mix the bacterial culture solution and gentian violet solution evenly, push the droplet away, and then heat the droplet with an alcohol lamp ( about 5 seconds) and let it dry naturally (about 5 minutes).
By direct observation with a 600x microscope, you can see a variety of bacteria and other microorganisms stained dark blue. However, the microorganisms seen at this time are all dead and immobilized, so their active state cannot be seen.
Prepare post-wash control mounts.
1. Wash your hands
Wash your hands with soap and dry them with a clean, dry towel;
2. Collect bacterial culture fluid
Rinse your hands with a small amount of sterile water and allow the hand sanitizer to flow into the clean petri dish;
3. Preparation of bacterial slices
The production process is the same as the film loading process above, just refer to the production process.
