Basic operation steps of microscope and digital camera

 

1. Warm up the fluorescent light source. (for fluorescence microscopy)


2. When no slide is placed, adjust the shooting lever to the digital camera for white balance.


3. Adjust the shooting lever to the observation position of the eyepiece.


4. Adjust pupillary distance


5. Adjust parallax


6. Find the target in bright field and low magnification objective lens, focus


7. Change the appropriate objective lens and focus.


8. Adjust the brightness of the light source


9. Adjust the condenser


10. Adjust the filter or aperture as needed


When a clear image is visible in the eyepiece, adjust the microscope to the digital camera as follows:
11. Adjust the shooting lever to transfer the image to the digital camera.


12. Check whether the focus is in focus under bright field of view, if not in focus, turn the fine focus knob to focus. At this time, the bright field should be used, and the camera should be kept at a high frame rate.


13. To adjust the color of the camera, you can do manual white balance as needed, or manually adjust the gain values of red, green, and blue.


14. Take pictures,
When shooting, in the bright field of view, the cmos camera should lower the gain value and adjust the saturation as needed.


When taking fluorescent photos, you should choose an appropriate gain value according to the recommendation, and lower the frame rate of the camera to ensure that the noise of the photos taken is low.


If you need to take better fluorescence pictures, the camera should be optimized for microscopic photography, and can be cooled to reduce noise. In addition, the best frame rate of the camera can be adjusted to reduce noise.
