Confocal Microscope Precautions and Maintenance

 

Confocal microscopy can measure a wide variety of samples. These include biological materials, tissues (sections), cellular (subcellular) structures and so on. The sources of fluorescence in the samples are as follows: autofluorescence, fluorescence staining, fluorescence, fluorescent protein, induced fluorescence (induced fluorescence) and enzyme fluorescence (enzymatically produced fluorescence) and so on. The excitation and emission wavelengths of most of these fluoresceins can be found in the dye information library of the software that comes with the instrument.


Confocal microscope instrument parameters
Laser:
UV laser: 405 nm, power > 30 mW;
Ar ion laser: 458/476/488/496/514 nm, power > 50 mW;
White laser: pulsed laser, 470-670 nm, power ≥ 1 mW, step 1 nm, 8 excitation spectral lines can be used at the same time, pulse frequency 80 MHz-10 MHz adjustable.


Objective lens:
Flat field complex achromatic confocal special fluorescence objective lens.


Detector:
Two PMT and HyD detectors each, one of which is HyD SMD detector with single molecule detection function.


Image Acquisition Processing:
LAS X software (preview, crosstalk separation, jigsaw, multipoint acquisition, data analysis), SymPho Time 64 (lifetime imaging, data fitting analysis), Huygens Professional (image deconvolution processing, visualisation and analysis, etc.).


Live cell workstation:
Temperature controlled + CO2 premixing.


Confocal microscope precautions and maintenance
Keep away from sources of electromagnetic radiation around the instrument;
The environment is free from vibration and strong air disturbance;
The chamber has a shading system to ensure that the fluorescent samples will not be bleached by exogenous light;
Clean environment;
Control the working temperature at 5~25℃.
