How does the microscope adjust the central image and phase contrast ring device?

 

1. On the basis of the adjusted Kuhler illumination system, use the bright field method to focus the sample clearly;


2. Turn the condenser to Ph1 to align with the scale line of the turntable, select a 10× phase contrast objective lens, and replace it with a transparent sample to be observed;


3. Unplug one of the eyepieces, replace the centering telescope, and focus on the two phase contrast rings in the field of view (the black phase contrast ring of the objective lens and the light transmission phase contrast ring of the condenser);


4. The two phase difference rings in the field of view do not necessarily coincide. Adjust the two adjustment devices on the condenser (the adjustment lever for adjusting the left and right positions of the phase difference ring and the friction knob for adjusting the front and rear positions) to make the light transmission ring move back and forth, left and right coincides with the black ring;


5. After adjustment, switch back to the observation eyepiece, press the green filter into the light path, and then observe the phase contrast image of the sample;


6. When observing with 20× and 40× objective lens, the condenser lens should be set at the Ph2 position, and when the 100 objective lens is used, the condenser lens should be set at the Ph3 position.
