Optical microscope mainly consists of objective lens group and eyepiece, which are usually equipped with lens groups with different magnification. Through combination, a very large-span magnification range can be formed. This configuration is because although the details of the high-power lens group are clearly displayed, the field of view and depth of field are narrow and it is not convenient to move in different target areas. Although the magnification of the low-power lens group is small, the field of view and depth of field are large, which is convenient for searching for targets in a wide range. Besides, some specific samples do not need large magnification, but all the targets in the field of vision are required to be as clear as possible, so the low-power lens group is also useful. The combination of the two can obtain perfect and clear imaging.
Required materials and tools:
Objective lens group: such as 100x, 200x, 300x, 600x, etc.
Eyepiece group: such as 5x, 10x, 15x, 20x, etc.
Focusing handwheel: including coarse adjustment and fine adjustment.
Stage: it can move in any direction in a plane, which is convenient for finding the target area.

Step method:
Turn on the light source
Rotate the coarse adjustment handwheel to pull the objective lens group away from the objective table to a safe distance.
According to experience, change into low-power objective lens group and appropriate eyepiece lens group.
Put the necessary processed samples on the loading table.
Adjust pupil distance of eyepiece
Move the sample through the stage, search for the sample and the target area, and observe and select the target through rough adjustment of the handwheel.
Replace the appropriate high-power objective lens and eyepiece, and focus carefully by fine adjusting the handwheel.
After clear imaging, research can be carried out and photos can be taken if necessary.
After finishing the work, turn off the light source and take down the sample. The objective lens group and eyepiece group must also be taken down and stored in a special dry and cool place, such as a drying bottle.
Precautions:
Skills of focusing: When fine-tuning focusing, there must be a process of blurring-clearing-re-blurring, in order to confirm the most accurate focus, so it is necessary to overdo it a little and then turn back to the clear focus position.
For some samples with specific shapes, color filters can be selected to improve the clarity of details. For example, a color filter with a specific color can be inserted into the optical path for a substance sensitive to a narrow wavelength and a sample dyed by fluorescence. In addition, in order to observe the special tissue in the metal sample, we can insert a polarizer, adjust the angle, and study the tissue condition by observing the specific polarized light reflected by it.
