How to Properly Use the Biological Microscope 100 Oil Lens

 

The specific operation steps are as follows.
(1) First find the specimen with a low-power objective, then switch to a high power objective and move the specimen to the center of the field of view, place the diaphragm on the zui, raise the lens barrel about 1.5 cm (or the carrier table down 1.5 cm), so that the objective lens is turned away from the optical axis.


(2) Remove the specimen slice and place 1 or 2 drops of cedar oil on the lens plane at the upper end of the condenser lens, then return the specimen slice to its original position so that the underside of the slide is in close contact with the cedar oil, to prevent gas saturation in the oil. However, this step may be omitted in the usual operation.


(3) On the top of the coverslip or on the top of the part of the smear specimen required to observe 1 drop of cedar oil, convert the oil mirror to the optical axis, adjust the coarse focusing screw with particular care, so that the lens is slowly lowered and the distance between the front end of the oil mirror and the specimen is carefully observed, and the lowering of the oil mirror stops when the front end of the mirror begins to touch the drop of oil. This operation process must be very careful to prevent the oil lens crushed specimen pieces and damage to the oil mirror accident.


(4) Observe with the left eye from the eyepiece, use the right hand to adjust the micro-focusing spiral, so that the lens slowly focus to the correct working distance to see the specimen clearly. Please note: coarse, microfocus spiral do not get it wrong, coverslip should be thin, too thick when you can not focus, otherwise easy to crush the specimen piece and damage to the lens.


(5) Adjust the size of the diaphragm, so that the numerical aperture of the condenser lens with the oil mirror to obtain a clear image.''


(6) Observation is completed, timely and good cleaning work. First make the oil mirror rose to and specimen 1.5 cm apart, the oil mirror will be turned away from the optical axis, with a dry mirror paper gently absorb the oil on the oil lens, and then wet xylene-soaked mirror paper wiped twice, zui later with a


Clean mirror paper and then gently wipe 2 or 3 times. The oil droplets on the condenser lens should be wiped off in the same way. Specimen piece of cedar oil can be used to pull the paper method wipe clean, that is, with a small piece of mirror paper covered in specimen piece of oil droplets on the silt and then drop some xylene on the paper, while wet, the paper will be pulled outward, so that continuous for 3 or 4 times, both can wipe clean and does not damage the smear specimen. Precautions for the use of oil mirror:
(1) temporarily interrupt the use of oil mirror, the barrel should be raised, so that the objective lens turned away from the optical axis, in order to prevent the barrel sliding down to crush the specimen piece, damage to the condenser lens and oil mirror accident.


(2) After use, wash off the cedar oil in time, because the cedar oil is easy to stick dust in the air, and the lens may be abraded when it is scrubbed again after sticking dust. In addition, cedar oil is not easy to wipe clean after drying in the air, and it is corrosive to the adhesive joints of the lens over time.


Mildew, gray fog and other hazards, so it must be timely and thoroughly wiped.
(3) When using oil mirror, generally do not carry out the same height focusing operation, because the same height focusing only applies to the original objective lens of each microscope, if the thickness of the specimen slice is too thick, when carrying out the same height focusing, the conversion oil mirror may touch the slide.


In addition to cedar oil as an oil immersion medium, water can also be used as a medium for the observation of living specimens in water, the operation method is basically the same as above.
