How to use the 100x objective lens of an optical microscope


Instructions:


(1) Access and placement When using the microscope, first take out the microscope from the mirror box. You must hold the mirror arm with one hand and the mirror base with the other hand to keep the mirror body upright. Do not use one hand to tilt and carry it to prevent the eyepiece from falling. To handle with care, put the mirror arm towards you, 5-10 cm away from the edge of the table. The table is required to be balanced, the table top clean and out of direct sunlight.


(2) Turn on the light source and turn on the power switch


(3) Placing the slide specimen Place the slide specimen to be microscopically examined on the stage with the material facing the center of the clear hole. Then clamp the two ends of the slide with spring clips to prevent the slide specimen from moving. If it is a slide mover, snap the slide specimen into the slide (4) mover, and then adjust the slide mover to move the material to the position facing the center of the clear hole.


(5) Observation with low magnification objective lens When observing the specimen with a microscope, first find the object image with a low magnification objective lens. Because the low magnification objective lens has a large observation range, it is easier to find the object image, and it is easy to find the part that needs to be carefully observed. The law is as follows:


1. Turn the coarse adjustment screw, look from the side with your eyes, and lower the lens barrel until the low magnification objective lens is about 0.5 cm away from the specimen.


2. Use the left eye to observe from the eyepiece, open the right eye naturally, and slowly turn the coarse adjustment screw by hand, so that the lens barrel gradually rises until the image in the field of view is clear. After that, use the fine-tuning screw to adjust the focal length slightly to make the object image the clearest.


3. Gently move the slide or adjust the slide mover by hand to find the part you want to observe. It should be noted that the object image in the field of view is an inverted image, and the slide should be moved in the opposite direction.


(6) High magnification observation On the basis of low magnification observation, if the magnification is not enough, high magnification observation can be carried out. The method is as follows:


1. Move the part to be observed to the center of the field of view of the low magnification lens, and the object image should be clear.


2. Rotate the nosepiece to move the high-power objective to the correct position, and then adjust the micro-spiral slightly to make the object image clear. This is due to the co-height focusing of the objective. If the microscope cannot be adjusted to the same height, or if you start to use a certain microscope and you are not familiar with its performance, you can operate as follows: after completing the low magnification observation, slightly raise the lens barrel, and switch the high magnification objective lens to work position, and then looking from the side, carefully and slowly raise the objective to find the target. This prevents the lens from being damaged by contacting the objective with the specimen or by getting water or chemicals next to the temporary slide.


3. Gently move the slide specimen or adjust the slide mover to find the part to be observed carefully.


When using a high magnification objective lens, due to the close distance between the objective lens and the specimen, special care must be taken, and the coarse adjustment screw cannot be used, but only the fine adjustment screw.


(7) Change the slide After the observation, if you need to use another slide, turn the objective lens back to low magnification, take out the slide, replace it with a new slide, and adjust the focus slightly to observe. Never change the film under the high magnification objective lens to prevent damage to the lens.


Arrangement After using the microscope, raise the lens barrel, remove the slide specimen, clean the microscope, turn the objective lens away from the clear aperture to form an "eight" shape, and then lower the lens barrel to an appropriate height. If there is a slide mover, move it so that the objective lens does not touch. Finally, put the microscope back in the designated position and put on the special cloth bag.
