Key Do's and Don'ts for Microscope Use
※ Objective lenses mainly include 4X, 10X, 40X and 100X oil immersion models. The operating rule is to proceed from low to high magnification. When observing tissue sections, start with the 4X objective (delivering 40× total magnification when paired with a 10× eyepiece). Adjust the coarse adjustment knob until the section image comes into view; the light intensity may also be adjusted at this stage. Then switch to the 10X objective (100× total magnification), and use the fine adjustment knob to bring the image into sharp focus.
Condenser and Iris Diaphragm
※ A condenser is installed directly below the circular aperture of the stage, used to focus and regulate light. An iris diaphragm is attached beneath the condenser, and its control lever adjusts the aperture opening. In coordination with the light source, it determines the intensity of light passing through the condenser and striking the back of the slide. Excessive brightness often blurs the specimen due to insufficient brightness contrast. In this case, narrow the iris diaphragm to facilitate observation.
100X Oil Immersion Objective Operation
※ When using the 100X objective (1000× total magnification), first rotate the objective turret to move the current lens aside. Place a small drop of immersion oil on the coverslip - the immersion oil has a refractive index close to that of glass to prevent light scattering, and acts as an intermediate medium for clearer observation. Then carefully rotate the 100X objective into position until it gently contacts the coverslip. Since the gap between the lens and coverslip is extremely narrow, take special care during focusing to avoid the lens pressing against the coverslip, which may cause the slide to crack.
Focusing Procedure for Oil Immersion Lens
※ When using the oil immersion objective (1000× total magnification), first use the fine adjustment knob to raise the stage until the coverslip makes gentle contact with the objective. Then slowly lower the stage with the fine adjustment knob to focus downward and locate the focal plane. If the objective, immersion oil and coverslip become separated, repeat step 9 to restart the process.
Post-Use Cleaning for Oil Immersion Lens
※ After using the oil immersion objective, wipe the lens repeatedly with lens paper moistened with 95% or 99.5% alcohol, then wipe it thoroughly with clean, dry lens paper.
Post-Use Shutdown
※ After operation, lower the stage to its lowest position, turn off the light source, and fit the microscope with its dust cover.
