Operating procedures and precautions for inverted microscopes
Operating Procedure
1. Turn on the power.
2 . Brightness adjustment. Adjust the brightness adjuster on the left side of the mirror base to 12V.
3 . Light path setting. Set the light path adjuster on the right side of the mirror base to A .
4 . Select the NCB11 filter. For phase contrast microscopy, use a green filter.
5 . Set the field diaphragm and the aperture diaphragm of the condenser to *.
6 . The 10 × objective lens is moved into the optical path.
7 . The sample is placed on the carrier stage.
8. The condenser is centred. Narrow the field diaphragm until its edge can be seen through the eyepiece, turn the condenser adjustment knob to focus the field diaphragm on the surface of the specimen, aligning the centre of the field diaphragm with the centre of the field of view; .
9 . 40 × objective lens spotter adjustment centre. Turn to the 40 × objective and adjust the field of view diaphragm so that the outer tangent field of view. Adjust the condenser so that the centre of the field of view diaphragm coincides with the centre of the field of view; .
10 . Select the appropriate objective lens for microscopic observation; .
11 . Convert to phase contrast objective. Turn the converter so that the *small magnification phase contrast objective enters the optical path. Focus with the coarse/fine adjuster
12 . The phase contrast diaphragm is moved into the optical path. The ring diaphragm must be matched to the phase contrast objective.
13. Ring diaphragm is centred. Open the field of view diaphragm to * large, remove one eyepiece and replace it with a centring telescope, turn the centring telescope to focus the phase plate of the objective lens and the annular diaphragm of the condenser, so that the phase plate and the annular diaphragm centre coincide;.
14 . Perform microscopic observation with a suitable phase contrast objective. The annular diaphragm pattern must be matched to the objective, and each time the annular diaphragm is changed, it should be centred according to step 13.
Note
1. Do not touch the lampshade, and do not place heat-sensitive or volatile substances near the lampshade; 2.
2. Lens cleaning. Dust * good with a soft brush or gauze **, for fingerprints, oil and other difficult to remove marks, can be cotton, lens paper or gauze dipped in a little anhydrous methanol or ethanol **, * good with petroleum ether scrubbing oil immersion oil on the oil mirror. After removing the oil with anhydrous methanol or ethanol wipe the lens, if there is no petroleum ether, can also be used methanol compound rub a few more times (usually four times)
3. Do not use petroleum ether to clean the lens at the bottom of the eyepiece barrel or the prism surface of the eyepiece barrel; anhydrous alcohol and petroleum ether are very volatile, when used and the beginning of the power supply should be very careful.
4. Do not use alcohol, ether and other organic solvents to clean painted parts, which will lead to discolouration; for stains, use gauze dipped in neutral detergent to gently scrub; and
5. When not in use, cover it with a flannel and put it in a dry place, keep the objective lens and eyepiece in a desiccator.
6 . After observation, do a good job of cleaning up hygiene, registration of the instrument
