Precautions for Capturing Microscopic Images with Nikon Microscopes
To obtain high-quality microscopic images or micrographs with a Nikon microscope, you must follow standard operating procedures. The key points listed below are essential:
① When using a Nikon microscope, check the specimen carefully: confirm whether the section thickness is appropriate and whether the coverslip meets national standards.
② The objective turret of a Nikon microscope generally has 6–8 positions. Plan achromatic objectives are commonly used. If an objective with too low magnification is selected, the image quality will degrade. Therefore, it is recommended to choose a plan objective of a suitable magnification.
③ Adjust the condenser aperture diaphragm to match the numerical aperture of the objective lens, so as to achieve optimal image resolution.
④ The focusing system is an important mechanical component, and proper focusing operation is critical. For beginners or new operators of the microscope, skilled use of the focusing system enables fast image finding and focusing, improving work efficiency.
⑤ When slicing specimens on the stage, it usually moves in the X and Y directions. The double-layer mechanical translation stage features high precision and good stability, which can deliver good image quality when observing microscopic specimens.
⑥ Aplanatic condensers provide highly uniform illumination, and the observed microscopic images show excellent contrast.
⑦ The field diaphragm should be adjusted to an appropriate opening according to different specimens and objective magnifications. Excessively large or small openings will affect the imaging effect.
⑧ If the illumination system adopts a light source with low temperature and high color temperature, the observed microscopic images feature high resolution and sharpness.
