Leica Upright Fluorescence Microscope Operating Procedures

 

1. Connect the power supply, turn on the computer host and microscope power, and plug in the encryption dog;


2. Release the pin of the microscope eyepiece (only need to be pulled out halfway); The various settings of the microscope have been adjusted, and no one is allowed to change them without the consent of the manager;


3. Pathological images can be observed under a microscope; It is necessary to gradually switch from a low magnification lens to a high magnification lens (10x100 is an oil lens, which cannot be used with ordinary HE and immunohistochemical films). Rotate the left knob of the microscope to adjust the macro, and the right knob to adjust the orientation;


4. On the computer desktop, double-click "WinV2" to enter the image acquisition interface - click "Image" - select "Capture" - select "Preview";


5. Adjust the light intensity as needed, i.e. "INT" on the left side of the microscope, to achieve the best image quality. Other settings must not be changed arbitrarily;


6. After adjusting, click "Capture Image" to obtain the image, and then save it;


7. After use, log out of the system - Turn off the computer - Place the microscope in a location without a lens - Push in the eyepiece pin - Turn off the microscope power - Remove the encryption dog - Cover the instrument cover - Turn off the main power.


Precautions for Leica upright fluorescence microscope:
1. The room where the instruments are placed must be regularly ventilated to prevent the instruments from getting moldy;


2. Without the consent of the person in charge, data cannot be copied without authorization, and must be copied using formatted USB drives;


3. After using the instrument, please register it in the trial record book;


4. Please take away the waste when leaving and do not leave it in the instrument room.


Leica upright fluorescence microscope operation steps:
1. Connect the power supply, open the computer host, microscope, and fluorescence box, and plug in the encryption dog. Microscopic operation is the same as the Leica cell image analysis system;


2. Press the "CHANGECHBE" button on the right side of the microscope to convert fluorescence;


3. Open the desktop "Leica Application suite V3.3.0" to enter the image acquisition interface;


4. Select CY3 (red fluorescence), D (ultraviolet narrow spectrum), DAP (ultraviolet broad spectrum), and GFP (green fluorescence) according to the required fluorescence wavelength. Specific information can be found in the spectrum: Settings - Microscope - ILTurretIL TURRET (5-POS);

 

5. Click on "Camera" to adjust "Get Image" - "Save" as needed;


6. After setting up, click "Options" - "Get Image" - "Save";


7. After taking photos, exit the program first, then turn off the microscope, fluorescence box, and host, and cut off the power.


8. Precautions are the same as the Leica Cell Image Analysis System.
