Operation Instructions for Fluorescence Microscopes

① Switch on the light source. The ultra-high-pressure mercury lamp requires several minutes of warm-up to reach full brightness.

② For transmitted-light fluorescence microscopes, install the required excitation filter between the light source and the condenser, and mount the corresponding barrier filter behind the objective lens. For epifluorescence microscopes, insert the filter block containing the required excitation filter, dichroic beamsplitter and barrier filter into the optical path slot.

③ Observe with a low-power objective. Align the light source center according to the adjustment mechanism of the specific fluorescence microscope model, so that the light source is positioned at the center of the entire illumination spot.

④ Place the specimen slide and focus for observation. Precautions during operation:

Do not observe directly with the naked eye when no filter is installed, so as to avoid eye injury.

Special non-fluorescent immersion oil must be used when observing specimens with an oil immersion objective.

The high-pressure mercury lamp cannot be restarted immediately after shutdown. A 5-minute interval is required before re-ignition; otherwise, operation will be unstable and the service life of the mercury lamp will be shortened.

Observation Under the demonstration-stage fluorescence microscope fitted with a blue-violet excitation filter, cells stained with 0.01% acridine orange fluorescent dye are visible. The cell nucleus and cytoplasm emit fluorescence in two distinct colors upon excitation: dark green and orange-red.
