Protocols for the use of optical microscopes

(iii) Turn the objective converter so that the low-magnification lens is facing the light-through hole on the carrier table. First, adjust the lens to about 1 to 2cm from the carrier stage, and then gaze into the eyepiece with the left eye, then adjust the height of the condenser, adjust the aperture diaphragm to the maximum, so that the light is incident through the condenser to the lens barrel, which is bright in the field of view.


(iv) Place the slide to be observed on the carrier table so that the part of the slide to be observed is in the centre of the aperture through which the light passes, and then clip the slide with a specimen clip.


(v) Observe first with low magnification (objective 10X, eyepiece 10x). Before observation, turn the coarse focusing handwheel so that the carrier table rises and the objective lens gradually approaches the slide. Care should be taken so that the objective lens does not touch the slide to prevent the lens from crushing the slide. Then, the left eye looking into the eyepiece, while the right eye is not closed (to develop the habit of observing with both eyes open with a microscope, so that while observing with the right eye looking at the drawing), and turn the coarse focusing handwheel, so that the carrier stage slowly down, you can soon see the magnified image of the material in the slide.


(F) If the object image seen in the field of view does not meet the experimental requirements (the object image deviates from the field of view), you can slowly adjust the moving handle of the carrier stage. Adjustment should pay attention to the direction of the slide and the field of view to see the object image moving in the opposite direction. If the object image is not very clear, you can adjust the micro-focus handwheel until the object image is clear.


(G) if further use of high-magnification objective observation, should be converted to high-magnification objective lens before the object image of the need to magnify the part of the observation to the centre of the field of view (low-magnification objective lens into a high-magnification objective lens observation, the field of view of the object in the scope of the narrowing of a lot). (H) in the conversion of high-magnification objective and see the object image, according to the need to adjust the size of the aperture diaphragm or the height of the concentrator, so that the light meets the requirements (generally low-magnification objective lens into a high-magnification objective observation, the field of view to be a little darker, so you need to adjust the light intensity).


(ix) observation is completed, the objective lens should be removed from the light hole, and then the aperture diaphragm to the maximum, and then the carrier table slowly fall, and check the parts without damage (special attention should be paid to checking the objective lens is stained with water stained with oil, such as stained with water or oil to be wiped with a lens paper), check the processing can be boxed up after the completion of.
