Steps for observing bacteria under a microscope

 

Using bacteria from dirty hands to prepare tablets:
1. Collect bacteria from hands, rinse dirty hands with a small amount of sterile water (cold boiled water can be used instead), and let the water flow from washing hands into the culture dish. These dirty waters are bacterial culture media.


2. Prepare bacterial slides, take clean glass slides A and B, and drop a small drop of bacterial culture solution in the center of each slide; Then cover the A slide with a cover glass and observe directly under a 600x microscope.

(By using a slightly darker field of view and adjusting it properly, various bacteria and other microorganisms can be seen. It is easy to see active microorganisms, which is more attractive and educational for children.) If you want to further zoom in and be proficient in operation, you can move the observed object to the center of the field of view and then switch to a higher magnification lens for observation.


3. Drop a small drop of gentian violet staining solution onto the bacterial culture solution of B tablet, mix the bacterial culture solution and gentian violet solution with a toothpick, and push the droplet away. Then, heat the droplet with an alcohol lamp (about 5 seconds) and let it dry naturally (about 5 minutes).


By directly observing under a 600 fold microscope, various bacteria and other microorganisms that have been dyed deep blue can be seen. However, at this point, all the microorganisms seen have died and been fixed, so their activity status cannot be seen.


Prepare control tablets after washing hands.
1. Hand washing
Wash your hands with soap and wipe them dry with a clean dry towel;


2. Collect bacterial culture medium
Rinse hands with a small amount of sterile water and let the hand soap flow into a clean culture dish;


3. The process of bacterial loading and production is the same as the above loading process, and can be referred to as production.
