Compared with traditional optical microscopes, laser confocal microscopes have higher resolution, can observe multiple fluorescence at the same time, and can form clear three-dimensional images. In the observation of biological samples, laser confocal microscopes have the following advantages sex:


1. Continuous scanning of living cell tissue or cell slices can obtain fine three-dimensional images of cytoskeleton, chromosomes, organelles and cell membrane systems.


2. It can obtain images with higher contrast and high resolution than ordinary fluorescence microscopes, and at the same time has high sensitivity and excellent sample protection.


3. Acquisition of multi-dimensional images. Such as 7-dimensional image (XYZaλIt): xyt, xzt and xt scans, time series scans, rotation scans, area scans, spectral scans, and convenient for image processing.


4. Intracellular ionic fluorescent labeling. Single label or multiple labels, detect the ratio determination and dynamic changes of intracellular pH and sodium, calcium, magnesium and other ion concentrations.


5. Fluorescent labeling. Probe-labeled live cells or living cell biological substances of sliced specimens, membrane markers, substances, reactions, receptors or ligands, nucleic acids, etc.; multiple substances can be marked and observed simultaneously on the same sample.


6. No damage to cell detection, reliable and excellent repeatability; data images can be output in time or stored for a long time.


How to Solve the Trouble of Microscope Coarse Adjustment
The main failure of the coarse adjustment of the microscope is that the automatic sliding or lifting is inconsistent. Automatic sliding refers to the phenomenon that when the lens barrel, lens arm or stage is still at a certain position, it will automatically and slowly fall down under the action of the weight of the microscope itself without adjustment. The reason is that the gravity of the lens barrel, lens arm, and stage itself is greater than the static friction. The solution is to increase the static friction force to be greater than the gravity of the lens barrel or lens arm itself.


For the coarse adjustment mechanism of the inclined tube of the microscope and most binocular microscopes, when the mirror arm automatically slides down, you can hold the anti-slip wheel on the inside of the coarse adjustment handwheel with both hands, and tighten it clockwise with both hands to stop the slide. If there is no effect, you should find a professional to repair it.


The automatic slide of the microscope lens barrel is often mistaken for the loose fit between the gear and the rack. So add spacers under the rack. In this way, although the slide of the microscope lens barrel can be stopped temporarily, the gear and the rack are in an abnormal meshing state. As a result of the movement, both the gear and the rack are deformed. Especially when the mat is uneven, the deformation of the rack is more serious. As a result, some parts are bitten tightly and some parts are bitten loosely. Therefore, this method should not be adopted.


Due to the long-term disrepair of the coarse adjustment mechanism of the microscope, the lubricating oil is dry, and it will produce an uncomfortable feeling when lifting and lowering, and even the friction sound of the parts can be heard. At this time, the mechanical device can be disassembled for cleaning, greased and reassembled.
