The principle of laser confocal microscopy is that the light beam emitted by the LED light source is focused onto the surface of the sample after passing through a porous disk and objective lens. Afterwards, the light beam is reflected back to the measurement system through the surface of the sample. When passing through the pinhole on the MPD again, the reflected light will only retain the focused spot. Finally, the light beam is reflected by the beam splitter and imaged on the camera.
